循環器疾患の遺伝子治療に関する研究:新しい心血管用ベクター開発と臨床の検討

研究概要	申請者らは、高効率かつ安全な遺伝子導入ベクターとしてHVJ-liposome法に注目した。同法を用いて、血管、心臓への高効率な遺伝子あるいは核酸化合物導入などを報告してきた。HVJ-liposome法が、低い毒性でかつ効率のよい心血管用ベクターとして有用であることを示した。さらにHVJ-lipsome法を用い、循環器疾患に対する遺伝子治療戦略を開発した。心疾患における細胞移植治療の可能性を示し、NFkBデコイの同法による冠動脈からの導入が虚血性心疾患の有効な治療になりうることも報告した。さらにcdk 2 kinaseのアンチセンス移植心への同法による導入が移植後の血管肥厚を抑制することも示した。血管疾患に対しても、P21遺伝子の導入、AP-1デコイの導入、prostacyclin遺伝子の導入による再狭窄予防、さらにapolipoprotein(a)に対するrybozymeの導入による動脈硬化へのアプローチをなども報告した。また、NFkBデコイの再狭窄予防に対する有用性も検討した。これらの知見をもとに、E2Fデコイを用いた再狭窄予防のヒト臨床研究を既に実施しており、さらにNFkBデコイを用いたヒト再狭窄予防の臨床研究も申請している。これらのヒト臨床研究においてはベクターを用いないプロトコールであるが、さらに優れた効果を得るため、現在、HVJ-liposome法の臨床応用に向けてさらなる評価検討を行っている。
