歯周病原菌プロテアーゼを標的とした歯周病治療薬の設計と開発

研究概要	歯周病の主要な病原性細菌であるPorphyromonas gingivalisは2つのユニークなシステインプロテアーゼ[Arg-gingipain(RGP)およびLys-gingipain(KGP)]を産生している。これらの酵素はいずれも膜結合型および分泌型の両方で存在する。いずれの酵素も、細胞外にあっては、宿主蛋白質の分解を通じて歯周病原性を発揮するとともに、本菌の発育増殖に必須のアミノ酸・ペプチドやヘムの獲得に寄与している。一方、菌体にあっては、本菌の血球擬集活性やヘモグロビン結合活性を担うとともに、菌体表層蛋白質や分泌蛋白質の輸送やプロセシングに中心的な役割を果たしている。本研究は、こうしたRGPおよびKGPの歯周病原性に基づいて、これらを標的とする新たな治療薬の設計・開発を目的としたものである。昨年度の研究から、RGPによる蛋白基質のぺプチド結合切断の特異性に基づいて約10種類のオリゴペプチドが作製された。その中でトリペプチドから成るKYT-1はRGPに対しては最も強い阻害活性(IC_<50>=8.4x10^<-10>)を示し、哺乳動物由来のシステインプロテアーゼであるカテプシンB、LおよびHに対しては弱い阻害活性しか示さなかった。また、本菌の産生するKGPに対してもほとんど阻害活性を示さず、きわめて特異性の高い阻害剤であることが明らかとされた。また、KYT-1は、ヒトタイプIコラーゲン、タイプIVコラーゲン、免疫グロブリン(IgGやIgA)、およびα2-マクログロブリンなどのRGPによる分解を強く抑制した。一方、RGPは好中球のもつ食菌作用を強く阻害することが知られているが、KYT-1はこのRGPによる好中球傷害活性も強く抑制した。さらに、本研究では、P.gingivalisの培養上清がヒト歯肉線維芽細胞を強く傷害し、細胞死を誘導することを明らかにした。この細胞死はKYT-1を共存させることによって強く抑制されることも示された。これらの結果は、RGP遺伝子を欠損させた変異株(rgpA rgpB二重変異株やrgpA rgpB kgp三重変異株)を用いた実験結果とよく一致しており、RGPがもつ歯周病原性を明確に示したものである。したがって我々は、RGPの阻害剤として今回新規に開発したKYT-1が歯周病に対して有効な薬物となり得る可能性を示唆することができた。
