精子運動を制御する細胞情報伝達機構の生理化学的解明と分子遺伝学的解析

研究概要

精子運動開始機構;サケ科魚類精子運動開始15K蛋白質の再現性の高い精製法について、新たにリン酸化チロシン抗体を用い、15KDa蛋白質、更に45KDa蛋白の同定が可能となり、無担体電気泳動法を用いての精製が最終段階に到達した。精子運動活性化走化性機構;ホヤでは卵由来精子活性化走化性物質(SAAF)の分子構造を解明のため、大量な試料(卵海水)を採取し部分精製を行った。更に、精子走化性定量化indexをより精密に改良し、イオンチャネル阻害試薬、活性化試薬の影響について調べ、イオンチャネルの精子活性化走化性への関与を調べた。今年度は21kDa外腕ダイニン軽鎖の及び実体不明の26kDa蛋白質のリン酸化が精子活性化に決定的役割を明らかにしたので、予定のホヤカルモジュリン関連蛋白質の同定は行わず、この結果を論文としてまとめ、更にダイニン軽鎖のcDNAクローニングを開始した。クラゲは、今年度はエルニーニョのためか、採集が不首尾で研究が出来なかった。ほ乳類については人の卵子の使用が許可される見通しとなり、昨年度の卵胞液を使った研究に加え、2月からは、ようやく本格的に精子走化性を誘起する細胞情報伝達機構についての検討を開始出来る見通しとなった。ニシンではHSAPs結合蛋白質(受容体)が昨年度明らかとなったプロリルエンドペプチダーゼに加え3種(70K,80K,85K)あることが新たに明らかになり、それらのcDNAクローニングによる全構造の決定似向けての研究が進み出した。精子イオンチャネルに関する研究:ホヤ精子では人工膜に埋め込んだ膜画分を用いてイオンチャネルの測定を行った。ジマスではカリウムチャネルの分子クローニングを行っている。精子培養系;二枚貝の二型精子で、異型精子、正型精子で異なった蛋白質が発現していることが明らかとなり、今後それらの遺伝子発現の研究を進める。
