脊椎動物視細胞における光受容に関与するタンパク質の多様性

研究概要

組換えDNA技術、組織化学的方法、タンパク質機能・構造解析法を用いて、視細胞で光電気変換に関わるタンパク質の多様性を解析した。(1)視物質:カエルの紫感受性視物質及び緑桿体視物質、ヤモリの各種視物質をクローニングし系統関係を解析した。またメダカの視物質の吸収極大波長を求めた。(2)Sモジュリン類似タンパク質:カエル網膜から単離された2種のカルシウム桔合タンパク質、Sモジュリンは桿体に、s26は錐体に発現していることを、抗体を用いて明らかにした。大腸菌中でSモジュリン・s26のキメラタンパク質作り調べたところ、C末端付近のアミノ酸配列の違いにより、2つの膜結合能に従ってロドプシンのリン酸化能力も高くなることが明らかとなった。Sモジュリンのカルシウム結合の変異体(E86M、E121M)を作成して性質を調べると、E86Mはカルシウムイオンを1個しか結合せず、またロドプシンキナーゼも阻害しない。E121Mはカルシウムイオンを全く結合せず、膜結合性もロドプシンキナーゼの阻害活性もない。したがってSモジュリンは先ず、E121Mサイトにカルシウムイオンを結合し、E86Mサイトにカルシウムイオンが結合出来るようになり、またミリスチル基が露出して膜に結合出来るようになって、ロドプシンキナーゼを阻害できるようになると推定された。(3)アレスチン:各種脊椎動物の網膜cDNAからアレスチン様タンパク質のcDNA断片をクローン化した、それらの系統関係を調べると2つのグループに別れた。それぞれに属するメダカのcDNA全長をクローニングし、in-situ-hybridizationで調べたところ、1つが桿体に、他が4種類の錐体で発現していることが明らかになった。
(4)視物質キナーゼ:代表的脊椎動物の網膜cDNAを用いて視物質キナーゼcDNA断片を解析したところ、これまでに報告されていないサブグループが見つかった。
