C_3植物とC_4植物の相同組織の機能分化に関する比較形態学的研究

研究概要

反射電子像観察装置を取り付けた走査型電子顕微鏡を用いることによって、走査型電子顕微鏡レベルでの金コロイド免疫電子顕微鏡観察が可能となったが、本年度はさらに凍結割断法を組み入れることにより、Rubiscoなど葉緑体内部に存在する酵素タンパク質の局在性を観察することができるようになった。さらに、走査型電子顕微鏡下で葉緑体内のラメラ構造やミトコンドリア内部のクリステを観察することが可能となった。この手法により、C_4植物におけるRubiscoやPEPカルボキシラーゼの特異的分布を確認するとともに、C_3植物ではRubiscoが葉肉葉緑体と維管束鞘葉緑体の両方に分布することを観測した。また葉緑体表面に、抗アクチン抗体と反応するタンパク質を検出した。アルカリ処理により細胞内容物を消化除去した試料を走査型電子顕微鏡で観察し、C_3植物とC_4植物における維管束鞘細胞の細胞壁微細構造を比較した。いずれの植物においても、葉肉細胞との間の細胞壁には多数の原形質連絡が存在した。原形質連絡には、単独で存在するものとprimary pit-fildの中に集団になって存在するものがあった。原形質連絡の形状や密度には、C_3植物とC_4植物との間で明確な差異は認められず、C_3植物においても、維管束鞘と葉肉細胞との間で、C_4植物の場合と同様の活発なsymplasticな物資輸送が起こっていることが示唆された。イネ(C_3植物)の葉身を切り出し外部からショ糖を与えたところ、伸長中の葉身では維管束鞘葉緑体に多量のデンプンが蓄積したが、葉身展開後はほとんど蓄積せず、維管束鞘細胞のsinkとしての機能は齢によって変化することが明らかになった。多肉質C_4植物であるオカヒジキの諸細胞の発達過程におけるPEPカルボキシラーゼの分布を調べたところ、昨年度調べたRubiscoの場合と異なり、葉の成熟にともない表皮下1層目の細胞のみに蓄積することが明らかになった。
