枯草菌蛋白質分泌過程におけるSRP・SRP受容体系とSec分泌系との相互作用

研究概要

(1)枯草菌におけるα・アミラーゼ、プロテアーゼ、セルラーゼなどの有用酵素類を大量に分泌生産する過程でSRP様粒子がどのような働きをしているかを分子生物学的に明らかにするためにSRPの構造と機能を解析した。シグナル認識粒子は主にヒトやイヌの細胞で研究されてきた。ヒトSRPの構成成分であるSRP7SRNA、SRP54蛋白質の枯草菌での相同物質としてscRNA(small cytoplasmic RNA)、およびFfhが枯草菌の蛋白質分泌に必須であることを明らかにし、scRNAのドメインIV部分でFfhが結合していることが明らかになったので、さらにscRNAドメインI、II部分にはDNA結合蛋白質HBsuが結合しており、HBsuもまた蛋白質分泌に必須の因子であることが分かった。
(2)枯草菌における蛋白質分泌カスケードを解析するためにSRP様粒子、SRP様粒子構成成分Ffh、およびSecA蛋白質による分泌蛋白質前駆体の認識をin vitroで解析した。枯草菌FfhおよびSecA蛋白質はそのC末端に6xHisタグを導入して、発現・純化する。また分泌蛋白質融合体(5種類の異なる分泌蛋白質シグナルペプチド部分とβ-ラクタマーゼ完成型酵素部分との融合体)は大腸菌SecAts変異株を宿主として利用し、42℃で発現させ、純化した。これらをin vitroで反応させた後、SRP、Ffh、SecAと分泌蛋白質前駆体とが複合体を形成していることを免疫沈降法および化学架橋剤EDACによる方法で解析した。さらに分泌蛋白質前駆体の細胞膜へのtargetingと膜通過におけるFfh、SRP受容体、SecAの働きを解析し、分泌カスケードは前駆体はまずFfhと結合し、SecA・Ffh前駆体蛋白質の三者複合体を作って、膜を通過すると予想した。これらの結果を基礎に枯草菌における高い蛋白質分泌系がSRP-SRP受容体系とSec蛋白質系の共同作業によっていることを明らかにした。
