生理機能解析をめざした光学活性な有用生物活性物質の合成研究

研究概要	1.植物の生長の抑制と共にジャガイモの塊茎形成など分化誘導にも関わり、植物ホルモン様成長調節剤として認識されつつあるジャスモン酸誘導体であるツベロン酸メチル及びグルコシドの光学活性体を高純度で合成する事に成功した。現在生理試験に提供し、機能と構造の相関を追求するとともに、関連類縁対の合成も進行中であり、新機能の開発等応用面の開拓をも目指しつつある。細胞周期阻害物質の抗腫瘍性活性に着目して、新しいタイプの制癌剤としての可能性を持ち、細胞周期特異的阻害剤で分化誘導能を有するRadicicloおよびAcetonaphthalidineの合成及び機能解析のための類縁体合成、さらには結合蛋白精製への道を探った。その結果Radicicolの新規合成法を確立しつつあり、次年度までには完成予定である。また関連類縁体の中から活性のあるものを見いだしており、現在、結合タンパク発見単離を検討中である。Acetonaphthalidineの合成についてはつい最近合成に成功し、現在共同研究者が、生物学的研究を進行中である。細胞周期阻害作用と神経突起伸長作用を合わせ持つ興味深いepolactaene及び類縁体の合成研究を行い、骨格合成に成功した。 3.トリミンググルコシダーゼやマンノシダーゼの阻害剤として知られる低分子疑似糖化合物の新規合成法の開拓および誘導体化、改変による反応部位への取込み増強を目指し、強力な阻害活性をもつNectrisine及びepinojirimycin及び関連同族体の合成を行ない、類縁体を含めて、合成に成功した。活性の分離、選択性の増加などを共同研究者が探索中である。 4.植物毒素、Sorokinianinの合成にもすでに成功し、現在改良合成を進めている。
