微生物由来の細胞周期阻害剤の単離と作用機作に関する研究

研究概要

細胞は,遺伝子(DNA)を複製して2つの娘細胞へ分配するという一連の過程(細胞周期)を経て増える。細胞周期の調節機構が解明されるにしたがって、癌細胞とは,その制御機構に異常が生じて,無秩序に増殖する状態にあると理解されるようになった。こうした背景から,細胞周期を阻害する薬剤が新しい抗癌剤として期待されている。我々は本研究課題で、微生物代謝産物より新しい細胞周期阻害剤の探索を行い,細胞周期阻害剤の作用解析を行うことを目的とした。
【探索】細胞周期阻害剤の探索系には、マウス乳癌FM3A細胞から樹立したp34^<cdc2>キナーゼ温度感受性変異株tsFT210細胞を用いた。本細胞は、39℃でcdc2キナーゼの酵素活性が失活するためにG2期で細胞周期の進行を停止し、32℃に戻すと再び細胞周期の進行を開始した。この条件下で、微生物培養サンプルを細胞培養液に添加し、細胞周期の停止作用をフローサイトメーターで測定した。スクリーニングの結果、カビの1種Phoma属の株が新しいサイトカラシン類縁化合物RKS-1778を生産することを見出した。RKS-1778は,細胞周期をM期で停止させる。さらに糸状菌Aspergillus terreusが、新規なペプチド系化合物テルペプチンを生産することを明らかにした。
【作用解析】すでに我々が細胞周期阻害剤として見いだしていたホスミドシンの阻害作用を詳細に解析した結果,ホスミドシンはG1サイクリンの1つサイクリンD量を減少させることによってRBタンパク質のリン酸化を阻害し,細胞周期の進行をG1で停止させることを明らかにした。
