哺乳類卵巣に及ぼす一酸化窒素の作用とシグナル伝達の分子生物学的解析

研究概要	ブタ顆粒膜細胞の無血清培養系を用いて、[1]NO代謝物(亜硝酸塩)測定、[2]ゴナドトロピン受容体(LH-R)発現に及ぼすNO供与体あるいはNO捕集剤の影響、および[3]転写因子遺伝子の発現に及ぼすNOの影響、について検討した。NO測定については、反応試薬(硫酸+ヨウ化カリウム)に亜硝酸塩(標準品として亜硝酸カリウム)を加えることによって発生するNOガスを直接NO電極による計測を検討し、感度としては約10pmoLの信頼性の高い測定法を確立した。この方法によって、細胞分化の際に発生するNOガス(実際はその代謝物の亜硝酸塩)を測定したところ、分化誘導を開始して約40時間前後に急激なガスの発生が観察された。また、このガスが発生する10時間前にNOの捕集剤(carboxy-PTIO)を培地に添加するとゴナドトロピン受容体(LH-R)の発現(細胞膜上でのリガンドの結合試験)が抑制された。NO供与体(NOC18)を培地に添加した場合では、LH-Rの発現には余り大きな変化は見られなかったが、ゴナドトロピン刺激で発現が上がる転写因子の遺伝子の一つc-fosの発現がNO供与体の添加によって増加した。NO捕集剤処理では、転写因子遺伝子c-fosの発現は抑制されたが、恒常的に発現するATF-4にはNO供与体あるいはNO捕集剤処理によって変化は見られなかった。このことから、NOはプロテインキナーゼCの伝達系を変化させるモデュレーターの可能性が考えられ、今後の研究方向に重要な示唆を与えるものである。
