研究概要

カタラーゼは過酸化水素を水に分解するヘム酵素であり、生理的にはペルオキシソーム内で発生する過酸化水素を除去するペルオキシソームのマーカー酵素でもある。一方、過酸化水素はIC製品など精密機械の洗浄剤や数の子などの食品消毒剤として、将来的にその用途開発と大量需要が見込まれているが、発がん性などの毒性のため、環境への放出が危惧されている。メタノール資化性酵母C.Boidiniiが持っているカタラーゼ活性を利用して、菌体反応による過酸化水素の分解プロセスを開発するために、カタラーゼの菌体内高生産を目的として、ソーティングエンジニアリングにより、タンパクの発現場所についての検討を行った。そのために、カタラーゼのペルオキシソームへのソーティングシグナルを同定し、カルボキシル末端の-NKF配列がペルオキシソームへの輸送に必要であることがわかった。本来、カタラーゼはペルオキシソームに局在しているが、菌体反応における過酸化水素への反応性や細胞質でヘムが付加してペルオキシソームへ輸送されることを考えると、必ずしもペルオキシソームで生産することは有利ではない。そこで、カルボキシル末端の配列を除いて発現させると、カタラーゼは細胞質内でペルオキシソーム内よりも高い活性で、存在することがわかった。さらに、菌体反応として過酸化水素分解反応を行って反応効率が向上していることがわかった。
メタノール培養時、アルコールオキシダーゼ(AOD)とジヒドロキアセトンシンターゼ(DHAS)は、巨大なペルオキシソーム内の90%以上もの場所を占める酵素である。そこで、ペルオキシソーム内の空間(最大で細胞内空間の75%以上)を拡張して、異種タンパクを蓄積する場所として用いて、菌体内での空間を供給するために、AODとDHASの2重遺伝子破壊株を作製した。
