視床下部の体温調節機能モジュール仮説の検討

研究概要	恒温動物の体温は多くの効果器系を動員して調節される.視床下部特に視束前野は体温調節の最高位中枢として温度情報の"統合coor dination"を行い、特に局所加温で活動が変化する温度感受性ニューロンが重要であると漠然と考えられている.申請者らは最近視束前野は多入力-多出力系の統合部位としてではなく、個々の効果器反応を調節する独立な神経モジュールの集まりであるという仮説を提案した.本研究の目的はこの仮説を組織学的、電気生理学的に検証することである.本年度は非ふるえ熱産生反応について視束前野からの投射様式を解析した.実験はWistar系雄ラット(300-500g)62匹で行い、少なくとも3週間5^○Cの環境温、14:10h明暗の部屋で飼育し寒冷順化させた.ラットはウレタンで麻酔し(1.4g/Kg)、手術台に固定、肩甲骨の後5cmを切開して熱電対を褐色脂肪組織の下に入れ、温度(Tbat)を測定した.視束前野(PO)には熱極或いは薬物投与用のカニューレを、視床下部腹内側核(VMH)に電気刺激用の電極を埋め込んだ.直腸温(Tre)も熱電対で測定しTbatとともに連続記録した.先ず、VMHを電気刺激し(0.075-0.3mA,33Hz,0.5ms)、TbatがTreより明らかに先行して上昇することを確認後、POを41.5^○Cに加温しながら、再びVMHを電気刺激するとTbatの上昇は全く起こらなかった.同様に、VMH刺激に対するTbatの上昇はPOに興奮性のアミノ酸であるD,L-ホモシステイン酸を投与によっても抑えられた.この結果は視束前野の温度が上がると興奮する温感受性ニューロンが褐色脂肪組織に抑制性の信号を送ることを示唆する.実際、POとVMHの間で視床下部を切断するとTbatは上昇するので、POからは定常的に抑制性信号が送られていることを示唆する.さらにTbatの上昇は外側部(外側視床下部)のみの切断でも起こる.POからの信号は内側前脳束を下行するものと思われる.
