寒冷順化における肥満ラットの褐色脂肪組織の代謝調節

研究概要	寒冷環境下では体温維持のため熱産生量が増加し,エネルギー消費量、摂食量が増加する.ラットでは寒冷曝露により,主に褐色脂肪組織(BAT)の脱共役タンパク質UCP1が増加して非ふるえ熱産生量を増加させる.今までの実験から,Wistarラットを長期寒冷順化させ,その後,脱順化させると寒冷順化群(C群)は脱順化群(D群)、温暖順化群(W群)より体重あたりの代謝率が高く,W群においては10℃の呼吸商は25℃の値より大きくふるえ産熱が優位であるがC群、D群では逆の結果を得た。D群ではBATはあまり減少しておらず代謝率の減少はエネルギー産生ホルモンの分泌減少により調節されている事を示唆した.肥満の機構を解明するために,Zucker fattyラットに寒冷順化を行い,同様の実験を行ったところ,寒冷順化,脱順化に対するエネルギー代謝の変化はWistarラットとほぼ同じ傾向を示した.しかし,摂餌量は寒冷曝露で増加せず体重増加の抑制はより顕著で脱順化による体重回復も遅れた. 今回更に詳細に調べる為,leanラットにおいて同様の実験を行い,三者の各群の体重,摂餌量の変化及び10℃と25℃における酸素消費量(代謝量),BATの形状,重量,BAT,白色脂肪、筋肉組織、おけるUCPs(UCP1-3)とGLUT4の発現,、血清の各種ホルモン値,脂質量等を比較した.fattyラットはleanラットと比べ寒冷順化による酸素消費量の増加率が著しく大きく、摂餌量はほとんど増加せず、体重増加の抑制はより顕著で、脱順化による体重回復も遅れた。 BAT重量は約3倍であったがUCP1、GLUT4の発現量の変化はleanラットとほぼ同様であった。BATの組織像をみると脂肪含有量はfattyラットでは著しく多く,leanラットでは著しく少なく,leanラットは温暖群においても,他のラットより産熱量が多い事実と一致する.
