視床下部神経内分泌細胞を用いたGABA_B受容体の電気生理学、分子生物学的解析

研究概要	(1)ラットの視索上核(SON)のスライス標本において、スライスパッチクランプ法を用い、神経内分泌ニューロンの細胞体のシナプス電流に対するGABA_B受容体アゴニスト、アンタゴニストの効果を測定した。GABA_B受容体アゴニストであるbaclofenは可逆的に抑制性および興奮性シナプス後電流(IPSCsとEPSCs)の発現頻度を減少させた。baclofenはシナプス電流の振幅および減衰の時定数には影響を与えなかったことからその作用はシナプス前に限局したものであることが示唆された。またGABA_B受容体アンタゴニストである2OH-saclofenはシナプス電流の発現頻度を増加させたことから内在性のGABAによってGABA_B受容体が持続的に活性化している可能性が示唆された。 (2)スライス標本をさらに酵素処理して、単離SONニューロン標本を作製し、(1)と同様の装置でホールセルパッチクランプ法を用いて、膜電位依存性Ca電流に対するGABA_B受容体アゴニストならびにアンタゴニストの効果を測定した。baclofenは可逆的にCa電流を減少させ、その効果はGABA_B受容体アンタゴニストであるCGP35348ならびにCGP55845により拮抗された。 (1),(2)の結果から視索上核にはシナプス前、後ともにGABA_B受容体が存在し、機能調節に役割を果たしていることが示唆された。 (3)ラット脳からmRNAを抽出し、アフリカツメガエルの卵母細胞にCFTR(Cystic Fibrosis Transmembrane Conductance Regulator Gene)CIチャネルあるいはGIRK(Gprotein activated Inward Rectifying K channels)と共発現させ、GABA_B受容体発現のセンサーとして用いることにより、cAMP変化によるCI電流、あるいはGIRK電流を指標にGABA_B受容体発言を確認した。
