ホスフォイノシタイド代謝共役受容体シグナル伝達機構の分子薬理学的分析

研究概要	交感神経α受容体アゴニスト、エンドセリン(ET)およびアンジオテンシンII(Ang II)は、ともにGqタンパクを介してPLCβを活性化することによりホスフォイノシタイド(PI)代謝促進の結果イノシトール1,4,5-三リン酸(IP_3)とジアシルグリセロール(DAG)を生成する。これらの受容体アゴニストはPI代謝促進により種々の細胞内情報伝達過程を修飾する。今年度の研究では、主として以下の課題に焦点を絞って実験を施行した。(1)DAGはPKC活性化を介してNa-HおよびNa-Ca交換機構機構の活性化を起こすことが心筋細胞およびin vitroの実験系で示されているが、intactな細胞における調節における意義の詳細はまだ不明である。我々は選択的Na-H交換機構およびNa-Ca交換機構遮断薬であるEIPAおよびKB-R7943を使用してET-1およびAng IIのindo-1負荷ウサギ心室筋細胞に対する効果に対する影響から両交換機構の調節における重要性を検討した。これらのアゴニストの効果には交換機構の関与があることが明確に示された。(2)ウサギ心室筋細胞のET受容体は、選択的ET受容体遮断薬に対して他の哺乳類心筋のET受容体とは異なった薬理学的作用様態を示す。この原因としては種々の期序の関与が考えられる。その一つとして、ET_AおよびET_B受容体をクローニングしたが、そのアミノ酸組成の相同性は90%以上とアミノ酸配列から薬理反応様態の差異を説明するのは難しい。現在培養細胞に発現させて結合性状の差異さらに共役過程の差異を検討する実験が進行中である。(3)心筋細胞α1受容体サブタイプハ少なくとも三種類存在する。最近ウサギ心臓にα1D受容体のmRNAの存在が確認されたが、その心機能調節における意義は不明であった。選択的遮断薬BMY7378を用いてα1D受容体が心筋収縮機能調節に関与していることを明確に示した。
