腎特異性有機陰イオン薬物トランスポーターの遺伝子クローニングと機能発現

研究概要

生体に投与された薬物の多くは肝臓を中心とする薬物代謝酵素によって極性が付与されて水溶性代謝物に変換される。これら代謝薬物はプラスないしマイナスの荷電を有するが、それらの多くは有機陰イオンであり細胞膜の透過に際して特異的な輸送膜タンパク質、即ちトランスポーターが必要である。他方、薬物の体外排泄経路には肝臓から胆汁・便、腎臓から尿、の2つが主要である。本研究では腎に特異性の高い発現が認められる有機陰イオン物質のトランスポーターの分子クローニングとそれらの機能解析を行った。
最終年に当る平成11年度には有機アニオントランスポーター(OATと略す)の第3番目のアイソフォーム、OAT3及び第4のOAT4の分子クローニングに成功した。ラットOAT3は腎と肝、脳、目に発現し、536アミノ酸から成る12回膜貫通の糖タンパク質で、既にクローニングしたOAT1と49%、OAT2と39%のアミノ酸の相同性を示した。輸送基質(とKm値)は、パラアミノ馬尿酸(65μM)、オクラトキシンA(0.74μM)、エストロン硫酸(2.3μM)を始めとする多くの有機アニオンを認識することが明らかにされた。ヒトOAT4は腎と胎盤に強い発現を認め、550アミノ酸から成る12回膜貫通が予測される糖タンパク質で、OAT1〜3とは38〜44%のアミノ酸の相同性が認められた。輸送基質(とKm値)はエストロン硫酸(1.01μM)、デヒドロエピアンドロステロン硫酸(0.63μM)、オクラトキシンAとはじめ、数多くの有機アニオンを認識する所謂多選択性トランスポーターである事を明らかにした。
3年間の本研究で分子クローニングできたOAT1,OAT2,OAT3,OAT4は全てが化学構造の異なる数多くの有機アニオンを認識するが、その分子機構の解明は今後の研究に委ねられた。しかしOAT_Sは有機カチオントランスポーターファミリー並びにカルニチントランスポーターファミリーと共に共通の系統樹を構成することから両性イオンのカルニチンを輸送するトランスポーターからアニオンとカチオンのトランスポーターが分子進化したものと想定される。更にOAT_Sの基質認識はアニオン部分と疎水性部分の2点が重要でその他の化学構造には特異性が極めて低いために多選択性を示すものと考えられた。
