ウイルス感染におけるフリーラジカルの分子病態とそのレドックス制御についての研究

研究概要

我々はこれまでの研究により、マウスインフルエンザウイルス肺炎モデルにおいて、宿主の炎症反応に伴って過剰に産生されるスーパーオキサイド(O_2^-)と一酸化窒素(NO)の反応産物であるパーオキシナイトライト(ONOO^-)が、主要な病原因子として機能していることを明らかにしてきた。パーオキシナイトライトは、化学的反応性に富み、生体内において多彩な病態生理活性を発現することが報告されている。特に最近では、パーオキシナイトライトによるチロシンのニトロ化反応がアポトーシスの誘導に深く関わることが示唆されている。そこで、本年度はHPLC^-電気化学検出法を用いて、インフルエンザウイルス肺炎モデルにおけるニトロチロシン生成について解析した。あわせて、以前本モデルに治療効果を示したNO合成阻害剤やパーオキシナイトライト消去剤のニトロチロシン生成、および、ウイルス感染病巣におけるアポトーシス誘導に与える効果についても検討した。その結果、ウイルス感染肺組織のみに有意なニトロチロシン生成が認められた。さらに、このニトロチロシン生成は、NO合成阻害剤やパーオキシナイトライトに対し強い消去活性をもっているエブセレンを投与することにより、強く抑制されることがわかった。またあわせて、ウイルス感染肺におけるアポトーシスの誘導をtunel法によりin situで検出し、アポトーシスの発現に対するエブセレンの作用について検討したところ、アポトーシスの誘導が、エブセレン投与により強く抑制されていた。同時に、炎症細胞の浸潤に伴った既存肺胞構築の変化などの病理組織学的変化は、エブセレン投与により著明に改善しており、マウスの生存率も有意に改善していた。以上の知見は、インフルエンザウイルスによる劇症肺炎モデルの肺組織損傷に、パーオキシナイトライトが深く関わっていることを示唆している。
