新たに同定されたNKR-P1陽性TCRα/β陽性胸腺細胞の分化と機能の研究

研究概要	従来、NK-T細胞は骨髄由来のCD4,8陽性未熟胸腺細胞上に発現されているCD1叉はTL分子によって自律的に、正の選択を受けるとされてきた。今年度の研究で、胸腺内のNK-T細胞の分化に必須のコンポーネントとして、胸腺髄質の構造が重要であることを世界に先駆けて明らかにした。即ち、胸腺髄質上皮細胞に欠陥を持つaly/alyマウスでは、NK-T細胞の前駆細胞には異常が無いが、NK-T細胞が産生されないことが判明した。また、脾臓のNK-T細胞ポピュレーションもaly/alyマウスでは小さく、IL-4産生能も低下していることを明らかにした。これらの異常は、正常胸腺組織を移植することによって修復された。次に、卵白アルブミン(OVA)特異的、I-A^d拘束性抗原レセプター(TCR)トランスジェニックマウスを用いてNK-不細胞の産生を解析し、このマウスにも正常の割合のNK-T細胞が存在することを明らかにした。従来NK-T細胞の正の選択には、NK-T細胞上のVα14-Jα281レセプターが必須と云われてきたが、今回の発見はMHCクラスII拘束性でVα13を発現するNK-T細胞が正の選択を受けることを初めて明らかにしてものである。また、このTCRトランスジェニックマウスにOVAを投与すると、メインストリームのT細胞のみならず、NK-T細胞のクローン消去が誘導された。今後NK-T細胞の分化と生理的機能を考えていく上で、大きなインパクトを与える発見と考える。現在、ZAP-70ノックアウトマウスを用い、このマウスではNK細胞は産生されるが、NK-T細胞とT細胞が産生されないことを明らかにしつつある。この研究はNK-T細胞の細胞系列、さらに免疫系における進化学的位置を解明する上で極めて重要なものである。
