トランスジェニックマウスを用いた癌転移の分子機構の解析

研究概要	多発性内分泌腫瘍症(MEN)2A型変異RET遺伝子導入トランスジェニックマウスの作製を行った。コドン634のシステインをアルギニンに変異させたRET遺伝子cDNAをモロニー白血病ウイルスLTRに結合し、マウス受精卵に注入した。その結果、約20コピーの遺伝子が組み込まれたトランスジェニックマウスが確立された。導入遺伝子の発現は脳、唾液腺、心臓、肝臓、精巣に認めた。病理組織学的な解析を行ったところ、このトランスジェニックマウスではすでに4週齢において甲状腺のC細胞過形成あるいは甲状腺髄様癌の発生がみられ、6ケ月齢ですべてに髄様癌の発生を認めた。腫瘍発生に伴って血清カルシトニン値も上昇し、髄様癌を発生したマウスでは3ないし8倍に上昇していた。その他の病理学的変化としては約半数例に唾液腺癌あるいは乳癌の発生を認め、乳癌を有するマウスでは高率に肺転移が存在していた。変異RET蛋白は予想されるように腫瘍細胞内で二畳体が形成され、ShcおよびGrb2アダプター蛋白と複合体を形成していた。一方、脳、心臓、肝臓では変異RET蛋白が高レベルで発現していたが、腫瘍発生は認めなかった。予想に反し、これらの組織では変異RET蛋白の二畳体形成はみられず、そのチロシンリン酸化も有意に上昇していなかった。現在その原因について解析中である。
