炎症における遅廷型血管透過亢進の機構

研究概要	本年度はサイトカイン測定に適したウサギ胸腔の急性炎症をLPS注射によって起こし、この炎症について白血球浸潤と血管透過亢進の動態を観察し、過去に用いた皮膚のアレルギー炎症におけるそれらと比較し、本質的には同様の所見であることを確認した。とくに血管透過亢進は2相性を示し、即時型透過は抗ヒスタミン剤で抑制されるが、遅廷型透過は影響を受けないこと、好中球枯渇操作によっては逆に即時型透過は影響されず、遅廷型透過は消失することを認め、今回のプロジェクトにとってこの炎症モデルが適していることを確認した。この系において産生されているサイトカインの産生動態を観察すると、炎症の初期(1〜2時間)で検出されるのはTNFαおよびIL-8で、IL-1βはこれらに遅れて約6時間でピークに達する。また、IL-8は炎症6時間で再び小さなピークを作り2相性に産生されることが判明した。IL-1の抑制因子であるIL-1raは9時間でピークに達し96時間まで長時間産生される。この系においてTNFαおよびIL-1βの機能を阻止すると、IL-1βおよびIL-1raの産生はほとんど完全に抑制され、早期の白血球浸潤は強く阻止された。後期のそれは一部しか阻止されなかった。一方、IL-1、IL-8、TNFαとの阻止により後期の白血球の浸潤が強く阻止された。従って、炎症の好中球浸潤はTNFαおよびIL-8に依存した即時型浸潤と、IL-1、IL-8およびその他のサイトカインに依存した遅廷型浸潤の2相があると考えられる。また、遅廷型血管透過亢進はIL-8またはTNFαの阻止によりほとんど完全に阻止されたので、今後TNFαおよびIL-8の血管透過における関与の様式を探ることが重要になった。
