研究概要	Tsc2変具(Eker)ラットの腎癌細胞に特異的に発現している新規Erc遺伝子の発現制御機構腎癌細胞と正常腎組織を用い、本研究室でmodifiedしたRDA-cDNAサブトラクション法により腎癌に特異的に発現してくる未知の遺伝子(Expressed in Renal carcinoma;ERC gene命名)を単離した。腎癌細胞で強発現しているERCの転写制御領域の解析を行ってた。がん抑制遺伝子Tsc2の発現抑制機構 Tsc2遺伝子発現制御機構を明らかにするためプロモーター領域の解析を進めた。DNA修復系酵素であるエンドヌクレアーゼIIIをコードするOcts3遺伝子がhead-to-headの状態で近接している。Tsc2遺伝子側、Octs3遺伝子側、各々のプロモーター活性を検出しており、この領域がoverlapしていることを明らかにしてきた。その領域には2ヶ所のcEts結合配列が存在しており、cEts結合配列のTsc2及びOcts3遺伝子発現制御への関与についてさらに検討を進めた。日本人結節制硬化性患者におけるTSC1、TSC2遺伝子異常の解析日本人結節性硬化症(以下TSC)患者について、TSC1およびTSC2の遺伝子解析を行った。 TSC遺伝子mutant腫瘍細胞に発現している遺伝子の単離・同定両側に腎癌を発症した1匹のEker ratから、それぞれ細胞株を樹立し、LK9d(R),LK9d(L),と命名した。この2つの細胞株は、いくつかの点で性状を大きく異にしている。まず、培養に際し、collagen coated plateを用いれば、ともに培養可能であるが、non-collagen coated plateを用いた場合はLK9d(R)のみ培養可能で、LK9d(L)は培養できない。細胞形態についてもLK9d(R)は類円形でflatな細胞形態をとるのに対し、LK9d(L)は紡錘形形態をとる。増殖速度は、LK9d(R)のほうが速い。Southernblot法では、rat染色体5番に存在するp16/15領域が、LK9d(L)では欠失が見られないが、LK9d(R)では欠失していた。今回、LK9d(R),LK9d(L)の2者間のcDNAサブトラクション法により初期の腎癌で特異的に発現してくる興味ある遺伝子を単離した。
