赤痢菌の細胞侵入と拡散の分子機構

研究概要	赤痢菌の上皮細胞侵入は本菌の感染初期の最も重要な機能であり、その理解は感染を初期段階で阻止する手段を講じる上で重要であると考えられる。本菌はM細胞から取り込まれ、マクロファージに感染しさらに吸収上皮細胞に対して側底面側から選択的に侵入することが知られている。そこで本研究では、赤痢菌の上皮細胞侵入機構を明らかにする目的で、本菌の分泌するIpaB IpaC,IpaD蛋白のファゴサイトーシス誘発に必要な、Ipa蛋白と上皮細胞表層の相互作用、およびそれにより引き起こされる細胞内シグナル伝達機構を分子レベルで解析した。具体的には(i)赤痢菌の細胞侵入に必要なシグナルの同定、(ii)Ipa蛋白添加に誘導される細胞内チロシンリン酸化蛋白と細胞骨格蛋白の同定と細胞内局在の解析、(iii)赤痢菌細胞侵入に於ける新規蛋白の同定およびその貪食誘導に於ける役割、の三つを細胞生物学的ならびに生化学的手法を用いて解析をおこなった。その結果、(i)赤痢菌の上皮細胞侵入はRhoサブファミリーの一つRhoの活性に依存していること、即ちRhoを不活化および活性化した上皮細胞へ赤痢菌を各々感染させると前者は細胞侵入が阻害され後者ではそれが亢進した。(ii)赤痢菌の侵入に伴い生ずる上皮細胞内反応は細胞接着斑の機能亢進 (α5β1インテグリンの集積、細胞接着斑構成蛋白質の局在)を誘導し、それはRho活性を阻害すると減弱した。(iii)休止期の繊維芽細胞に赤痢菌から遊離したIpaB,IpaC,IpaDを添加すると、その細胞にRhoの活性化で認められる反応が惹起された。(iv)IpaCはα5β1とともにαmβ2インテグリンに対しても結合性を示し、マクロファージへの感染に重要なはたらきがあることが示唆された。
