感染におけるマトリックスメタロプロテアーゼの活性化と病態増悪機構

研究概要

これまで我々は、病原細菌の産生するいくつかのプロテアーゼが、マトリックスメタロプロテアーゼ(MMP)を限定分解し、特異的に活性化することを明らかにしてきた。MMPは、細胞外マトリックス(コラーゲンなど)の改変をもたらす酵素群であり、本酵素の過剰な活性化は強い組織破壊や血管系の破綻をもたらすことが知られている。そこで本年度の研究においては、A群レンサ球菌のプロテアーゼ毒素のヒト好中球プロコラゲナーゼ(MMP-8)に焦点をあて、劇症型A群レンサ球菌感染病態解析を行った。その結果、A群レンサ球菌由来のSPE-B/チオールプロテアーゼと菌体表層プロテアーゼが相乗的に作用して、マトリックスメタロプロテアーゼを活性化することが明らかとなった。本菌の産生する2つのプロテアーゼは、宿主のMMPの活性化を介して組織破壊を促進し、菌体の組織的侵入を容易にし、重要な病原因子として作用しているものと思われた。あわせて、細菌感染に伴った炎症反応によって生ずる一酸化窒素(NO)および活性酸素(スーパーオキサイド、O_2^-)の反応生成物であるパーオキシナイトライト(ONOO^-)によるMMP-8の活性化メカニズムの解析を行った。その結果、パーオキシナイトライトはMMP-8を活性化し、さらにその活性化はグルタチオンの添加により著明に増強された。この際の活性化はMMP-8とグルタチオンの不可逆的な結合によりもたらされていた。この知見は、細菌感染病巣において、病原体より産生されるプロテアーゼのみならず宿主側因子であるNOや活性酸素が、MMPの活性化を介して組織破壊を増強させ、細菌感染病態に深く関わっていることを示している。
