研究概要

主要組織適合遺伝子複合体(MHC)クラスI分子によって提示されるペプチドの産生に与るマルチ・サブユニット・タンパク質分解酵素であるプロテアソームの構造と機能を解析することにより以下の結果を得た。
1)IFN-γ 刺激によってプロテアソームにとりこまれるPSMB10(MECL1)サブユニットとMECL1サブユニットによって置換される PSMB7(Z)サブユニットをコードするマウス遺伝子をクローニングし,その構造を決定した.その結果,これらの遺伝子はともに8個のエクソンからなり,同一のエクソン・インドロン構造をもっていることが明らかになった.Psmb10遺伝子のプロモータ領域には2個のIFN-stimulated response elementsが存在した。これらのエレメントを欠落させるとIFN-γ誘導性が消失することをCAT assayを用いて確認した。また,PSMB10,PSMB7サブユニットをコードする遺伝子はいずれもMHC外でコードされており,前者は第8染色体のSntb2遺伝子,後者は第2染色体のGrp78遺伝子近傍に存在することが明らかとなった.
2)PA28アクテベータ複合体は20Sプロテアソームに結合することにより,MHCクラスI分子によって提示されるペプチドの産生を促進する機能をもっている.マウスのPA28 alpha,beta,gammaサブユニットをコードする遺伝子を単離しその構造を決定することにより,これら三つの遺伝子は類似したエクソン・イントロン構造をもっていること,alpha,betaサブユニットをコードする遺伝子は約6kb離れて,その3′-endを向き合うようにして第14染色体上に存在することなどが判明した.
