膵液中に存在する補体関連蛋白の意義とその膵疾患診断への臨床応用に関する研究

研究概要	本年度は、膵管上皮細胞株(PANC-1、MIA PaCa-2)を用いて、膵管上皮細胞からの補体成分(C3,C4,factorB)の産生のサイトカイン制御について主に検討した。我々の検討によると、C3とfactorB産生は、IL-1β、IL-6、TNF-αにより誘導され、C4はIFN-γにより誘導されることが明かとなった。さらに、IL-4はこれらの補体産生に対して抑制的に作用することも明かとなった。一方、補体活性化制御因子decay-accelerating factor(DAF)の膵管上皮細胞における発現の制御機能の基礎的検討を実施した。即ち、DAFの発現は、DAFのペプチド部位の発現とGPIアンカー部位の発現により規定されているが、これらの蛋白(DAF、GPIアンカー律速酵素PIG-A)発現のサイトカイン制御について検討したところ、次のことが明かとなった。膵管上皮細胞において、IL-4やTNF-αがDAFの発現を強く誘導したが、IL-4はDAFペプチドの発現は誘導するがPIG-Aの発現には影響をあたえない。しかしTNF-αはDAFペプチドおよびPIG-αの両者の発現を遺伝子レベルで強く誘導した。この結果は、GPIアンカー蛋白の発現制御機構の複雑性を示す初めての知見であり興味深い。おそらく、膵炎において、DAFの発現は複雑の分子レベルの制御を受けているのと考えられる。今回得られた知見をもとに、本年度(平成10年度)に計画している臨床材料を用いた検討にすすみたい。一方、DAF遺伝子欠損マウス(KOマウス)の作成を試みている。現在までにおDAF遺伝子の片方に変異を導入したES細胞を樹立し研究中である。ただ、基礎的検討の段階であるが、DAF遺伝子のKOは、致死的となる可能性があり、この点のさらなる検討が必要と考えられる。 GPIアンカー合成酵素PIG-Aに対するポリクローナル抗体の作成に成功しており、平成10年度では、蛋白レベルの解析がさらに可能となるものと考えられる。
