研究概要

本研究は、サーファクタント蛋白質SP-A及びSP-Dの肺胞細胞受容体を介する機能の分子機構を解明する目的で遂行された。平成9年度に得られた成果を下記に要約する。
1.SP-A及びSP-Dの各種変異体の作製
コレクチン・ファミリー蛋白質のC末端糖鎖認識領域(CRD)の受容体及び脂質結合部位を解析するために各種のSP-A/マンノース結合蛋白質(MBP)キメラ体、SP-A/SP-Dキメラ体を作製した。
2.SP-A及びSP-Dの脂質結合及び肺胞II型細胞受容体結合の責任領域の解析
各種のSP-A/SP-D及びSP-A/MBPキメラ体を用いて検討した結果、SP-AのCRD領域Glu^<195>からC末端までの部位がリン脂質(DPPC)と肺胞II型細胞との結合領域であり、SP-DのCys^<261>からC末端までの領域がホスファチジルイノシトール結合領域であることを明らかにした。
3.SP-Aの肺胞マクロファージ受容体を介する生体防御作用の解析
生体防御におけるSP-A関与の機構を解析する目的で、SP-Aとエンドトキシンの主成分リポ多糖(LPS)との結合及びマクロファージ系細胞株U937細胞におけるLPS刺激によるTNF-α発現に及ぼすSP-Aの効果について検討した。その結果、SP-AはR型LPSには結合するが,S型LPSには結合しないこと、及びSP-AはLPSによる免疫細胞の活性化に対し異なる作用をもつことを明らかにした。
4.肺胞II型細胞におけるSP-A結合蛋白質アネキシンの役割の解析
アネキシンIVは、Ca^<2+>依存的にSP-Aと結合することを明らかにした。SP-Aの結合部位はアネキシンIVのC末端側にあり、ドメイン-2はその結合を阻害し、ドメイン-1がCa^<2+>依存的にドメイン-2の結合阻害を除く調節領域になっていることを示した。
