肺サーファクタント蛋白質A及びDの受容体を介する機能調節の分子機構と肺病態の解析

研究概要

親水性の肺サーファクタント・アポ蛋白質(SP-AとSP-D)の細胞II型細胞膜受容体を介するリン脂質代謝調節の分子機構、SP-A結合蛋白質(アネキシンIV)の肺胞における生理的意義を探るために、SP-AおよびアネキシンIVの機能部位を解明し、また、生体防御におけるSP-Aの役割の解明を進め、以下の結果を得た。
1. SP-AとSP-Dは相同であるが、リン脂質、糖脂質結合特異性が異なり、SP-Aは、DPPC、GalCerと親和性を示し、肺胞II型細胞からのリン脂質分泌・取り込みを調節するのに対し、SP-DはPI、GluCerと親和性を示す。SP-AとSP-Dのキメラ体を用いた実験から、SP-AのGlu195〜C末端がSP-Aの機能発現に重要であることが判明した。更に、SP-DのPIおよびGluCerとの結合に必要な領域が異なり、Glu321〜C末端がPIとの結合に、一方、Cys261〜C末端がGluCerとの結合に重要であることが示された。
2. アネキシンIVのC-端側には、70残基の4回の繰り返しドメインを持ち、各ドメインにはCa^<2+>およびリン脂質結合部位が存在する。アネキシンIVおよびSP-AのCa^<2+>結合部位に変異を加えた点突然変異体を用いた検討で、アネキシンIVのドメイン3へのCa^<2+>結合およびSP-AのArg197が、アネキシンIV-SP-Aの結合に重要であることが示された。
3. SP-Aがリポ多糖(LPS)、特に、o-抗原多糖を欠いたrough LPS及びリピドAに結合することを見いだし,更に、o-抗原多糖を持つsmooth LPSによるマクロファージのサイトカイン産生(TNFαなど)がSP-Aによって抑えられ、smooth LPS受容体であるCD14とsmooth LPSの結合をSP-Aが阻害することもわかった。従って、気道感染による過剰反応をSP-Aが制御しうることが示された。
