研究概要

1,171個のアミノ酸からなるMG160の3箇所と,human trans-Golgi networkの18-35アミノ酸残基(TGN46)に対するポリクロナール抗体を作成し,ALS例と非ALS例の腰髄および中心前回のパラフィン切片を免疫染色した.(1)今回新たに染色性の良好な二種類(MG160の590-605アミノ酸残基とTGN46)の抗体を作成できた.(2)ヒト脊髄に対する染色性は,Gonatasらが作成した抗MG160抗体とほぼ同様であった.(3)ALS前角細胞では,この二種類の抗体でもGolgi装置のfragmentationが高頻度に認められ,さらにGolgi装置の数の減少や分布の異常も観察された.(4)ALS前角内の小型神経細胞および長期生存例の残存する前角神経細胞のGolgi装置は比較的よく保たれる傾向を示した.(5)Bunina小体はこの二種類の抗体でも陽性所見は得られなかった.(6)ALSの一次ニューロンである中心前回Betz細胞のGolgi装置の検討では,ALS例・非ALS例の中心前回のBetz細胞のGolgi装置は脊髄前角細胞と同様に顆粒状〜紐状に明瞭に染色された.非ALS10例ではGolgi装置のfragmentationの頻度は約0.6%と極めて少なかった.17ALS例では,症例毎に差はあるものの,全Betz細胞の約15%にfragmentationを認め,その出現頻度は脊髄前角細胞に比して軽度であった.今回の検討より,ALS例のBetz細胞でも脊髄前角細胞と同様にGolgi装置の異常が生じていることが確認できた.
