膵β細胞のインスリン分泌顆粒形成能が前駆体切断酵素Furinによって減弱する機序

研究概要	前駆体活性化酵素Furinは、PDGF,HB-EGFなどの増殖因子,インスリン受容体やHGF受容体c-Met,カドヘリンやインテグリンα3とα6などの接着因子等の前駆体を活性型に転換する。膵β細胞で発現しているproPTHrPとproTGFβはFurinの切断配列RXXRをもつ。著者は増殖の速い膵β細胞株でFurinが高発現することを見い出した。そこで膵β細胞株MIN6を用い、1)PTHrP,TGFβ,Furinの共発現、2)Furin阻害剤によるproPTHrPとproTGFβのプロセシング抑制、3)増殖の遅い(継代数17)MIN6-17と増殖の速い(継代数41)MIN6-41に対するPTHrPの分化作用(インスリン含量)と増殖作用(^3H-TdR取込み)作用、を検討した。 MIN6のインスリン含量は継代数と共に低下し、MIN6-41はMIN6-17の半分以下の含量を示した。逆に^3H-TdR取込みはMIN6-41の方がMIN6-17より約2倍高かった。FurinとPTHrPは継代数の増加と共に発現が増加したが、TGFβは発現が減少した。Furin阻害剤はproPTHrPとproTGFβのプロセシングを抑制した。PTHrPはMIN6-17のインスリン含量を増加させ、更にFurinとTGFβの発現を誘導した。しかしPTHrPはMIN6-41で^3H-TdR取込みをより増加させ、PTHrP自身とFurinの発現を誘導した。従って、PTHrPとTGFβは各々の発現が上昇する条件でFurinと共発現した。PTHrPはMIN6に対して分化度の高低に関係なくFurinを誘導し、分化度の高いMIN6ではTGFβを高発現させて分化機能を維持すると考えられる。
