動脈硬化発症過程における血管壁へのリポタンパク繋留とその制御因子に関する研究

研究概要	動脈硬化の発症過程に3けろリポタンパク繋留反応仮説の妥当性を検証するために、アポリポプロテイン(アポ)Eのリポタンパク繋留に及ぼす作用を検討した。アポEを過剰に発現するトランスジェニックマウス(ETg)とLDLレセプターノックアウトマウス(LDLRKO)に交配して、アポEを過剰に発現するLDLレセプターノックアウトマウス(ETg/LDLRKO)を作製した。ETg/LDLRKOとLDLRKOの脂質代謝ならびに動脈硬化についで検討した。ETg/LDLRKOの血清アポE値は増加し、その増加分はトランスジーンでコードされるラットアポEに由来するものであった。血中のnon-HDL分画のコレステロール値はLDLRKOに比較して抑制された。また、Etgを1コピーもつヘテロ接合体(Etg÷/-/LDLRKO)に比較して2コピーもつホモ接合体(Etg÷/+/LDLRKO)において、上記の変化は顕著であった。超遠心で分画したリポタンパクの中のアポタンパクの分布を調べると、VLDLのアポE含量増加と、LDLに含まれるアポB-100の含量の低下が観察された。ETg/LDLRKO由来とLDLRKO由来のVLDLを同一の遺伝型のマウスに静脈注射して血液中からの消失速度を比較すると、ETg/LDLRKO由来VLDLの代謝速度が亢進していることが明らかになった。高コレステロール食負荷後に大動脈のアテローム病変はは約1/2に抑制された。
