糖尿病発症に関与する膵β細胞のグルコース代謝シグナル伝達とその応答障害の解析

研究概要

ラット膵ラ氏島の初代培養細胞に転写因子CREBまたはCRE-BPlを遺伝子導入すると、CREBではグルコース応答性のインスリン遺伝子発現は抑制されたが、CRE-BPlでは促進された。また、膵β細胞由来細胞株HIT-T15にCREBおよびCREMを遺伝子導入すると、CREBではグルコース応答性のインスリン遺伝子発現は抑制されたが、CREMでは促進された。したがって、CRE(cAMP responsive element)に結合するCREB/ATFファミリー転写因子のなかでもCRE-BPlやCREMがグルコース応答性のインスリン遺伝子の発現に重要な役割を有している。
点突然変異を導入した機能解析の結果、CRE-BPlの活性化には69、71.73番目のスレオニン残基のリン酸化が重要であり、細胞内カルシウム濃度の上昇に伴いリン酸化が認められた。リン酸化されたCRE-BPlは、転写のco-activatorであるCBP/p300と相互作用し、転写を調節することを見出した。
一方、CREMは作用の異なるisoformがalternative sp1icingにより産生されるが、膵ラ氏島で発現する新たなisoform、CREMΔQl、CREMΔQ2、CREM-17、CREM-17Xを見出した。これらの機能解析から、CREMのP領域やQ領域が、基本転写囚子TBPやTAF130と結合し、転写を調節することを見出した。
