糖尿病発症に関与する膵β細胞のグルコース代謝シグナル伝達とその応答障害の解析

研究概要	膵β細胞に発現する転写因子CREM(cAMP response element modulator)には、alternative splicingによって産生される異なるisoformが存在するが、このうちインスリン遺伝子の転写の促進に作用するisoformであるCREMταのインスリン遺伝子に対する転写活性は、CREMα、ICERによって競合的に抑制される。CREMταの転写活性には、リン酸化に関与するP領域とグルタミンに富むQ領域が重要であるが、これらPQ領域とGAL4 DNA結合領域との融合蛋白を用いて、インスリン産生HIT-T15細胞におけるグルコースによるCREM転写活性能を検討したところ、グルコース濃度の上昇に応じて転写の活性化が認められた。次に、protein kinase A(PKA)によるリン酸化部位であるP領域の117番目のセリンをアラニンに置換したところ、CREM転写活性のグルコース濃度応答性は消失した。したがって、117番目のセリンのリン酸化を惹起する細胞内情報伝達系がグルコース応答性のCREM転写活性化に重要であると考えられた。このような細胞内情報伝達系としてcAMP濃度の調節が考えられる。膵β細胞内のcAMP濃度を調節する消化管由来のペプチドホルモンであるGIP(gastric inhibitory polepeptide)の生理活性に関して、受容体欠損マウスを作製して検討したところ、GIP受容体欠損マウスでは食直後のインスリン分泌が低下し、耐糖能が低下することを明らかにした。以上の結果から、cAMPを介する情報伝達系は、インスリン合成・分泌いずれにも大きな役割を有することが明らかとなった。
