研究概要	I. IL-1遺伝子に関する成績 1. HTLV-I非感染T細胞白血病株細胞であるJurkat細胞に、IL-1β遺伝子上流とともにHTV-I感染の産物であるTAX蛋白を挿入して(ATL細胞モデル)CATassayで測定したIL-1β遺伝子活性化はTAX蛋白非挿入群に比して著明に増加した。この際、IL-1β遺伝子上流に結合する転写活性因子の一つであるNFIL-6およびSp-1の結合部位を変異させたところ、TAX挿入群でも活性化は全く認められなかった。 2. 1と同様の手法でJurkat細胞に、IL-1α遺伝子上流とともTAX蛋白を挿入して測定したIL-1α遺伝子活性は著明に増加したが、IL-1α遺伝子上流に結合するNF-κBの結合部位を変異させたところ、TAX挿入群でも活性化は全く認められなかった。以上の結果より、IL-1の産生には、HTLV-I感染の産物であるTAX蛋白が強く関与し、Tax蛋白によるIL-1遺伝子の誘導にはIL-1遺伝子上流へのNFIL-6、Sp-1、NF-κB等の転写活性因子の結合が必須であることが判明した。 II. 接着分子に関する成績 1. 破骨細胞による骨吸収は骨芽細胞を介して惹起されることが知られている。ATL細胞と骨芽細胞株細胞であるHOS細胞とのheterotypicな接着によってHOS細胞へのIL-1βmRNAの発現および細胞からのIL-1β産生増強を認めた。 2. この発現・産生は細胞接着分子VLA-4,LFA-1等に対する抗体で著明に減少した。以上の結果より、IL-1の産生には複数の接着分子を介する骨芽細胞とATL細胞の接着も強く関与していることが判明した。
