脳虚血における神経栄養因子および神経伝達修飾物質の動態解析

研究概要

ラットの脳虚血モデルを用い、海馬や線条体での遅発性神経細胞死や大脳皮質の梗塞に関して、各種神経伝達修飾物質の動態や、脳保護物質の効果について検討した。さらに、マウス海馬CA1領域に発現する未知遺伝子の単離を行い、今後の研究の方向性を模索した。その結果判明した主な点を以下に述べる。
(1)虚血性遅発性神経細胞死はアポトーシスと関連が深い。
(2)一酸化窒素合成酵素阻害剤は線条体からのドパミン放出を抑制する。
(3)ダントロレンは海馬の遅発性神経細胞死を改善し、一酸化窒素やグルタミン酸の放出を抑制する。
(4)過酸化脂質代謝酵素のglutathione peroxidaseを脳室内投与しても遅発性神経細胞死は改善しない。
(5)静脈全身麻酔薬のプロポフォールは虚血海馬からのグルタミン酸放出を抑制し、遅発性神経細胞死を改善する。しかし、プロポフォールの神経細胞保護作用は、グルタミン酸放出抑制よりもむしろ抗酸化効果によるものである。
(6)アンチトロンビンIIIは脳梗塞巣の範囲を軽減する。
(7)遺伝子トラップ法により、マウス海馬CA1領域に発現する未知遺伝子の単離に成功した。今後その遺伝子機能を解析することで、脳虚血病態の解析や脳保護物質の開発を進める。
