トロンボキサンA_2受容体サブクラスとその細胞情報伝達機序の研究

研究概要	トロンボキサンA_2(TXA_2)受容体のサブクラスの発現について、mRNAレベルおよび蛋白質レベルで検討した。はじめに、アストロサイトーマ細胞(1321N1)に発現しているTXA_2受容体mRNAについて調べるために、placentaタイプおよびendothelialタイプのTXA_2受容体mRNAを同時に検出できるプライマーを設計し、RT-PCRによって増幅した。その結果、placentaタイプおよびendothelialタイプの二つのTXA_2受容体mRNAの存在を明らかにした。一方、アストロサイトーマ細胞より細胞膜を調製し、それを可溶化してTXA_2受容体の精製を試みた。すなわち、TXA_2受容体の部分ペプチドに対する抗体を結合させたイムノアフィニテイカラムを作製し、それを用いて、可溶化物からTXA_2受容体を含む画分を部分精製した。[^3H]SQ29548を用いた受容体結合実験によって、TXA_2受容体が濃縮されていることを確認した。さらに、TXA_2受容体抗体を用いたウエスタンブロッテイングにより、部分精製画分には55および58kDaに二つのバンドを検出した。すなわち、蛋白質レベルでにおいてもサブクラスの存在を明らかにした。このTXA_2受容体を含む部分精製画分には、G_<qα>抗体と反応するG蛋白質とともに、G_<12α>抗体と反応するG蛋白質が認められた。すなわち、ヒトアストロサイトーマ細胞には二つのTXA_2受容体サブタイプの存在することをmRNAおよび蛋白質レベルで明らかにすることに成功した。また、TXA_2の遊離を介して血小板凝集を引き起こすゾ-ザンテラトキシン-Aの作用解析から、TXA_2受容体刺激はmitogen-activated protein kisanse(MAPK)のリン酸化化をもたらす細胞情報伝達系を活性化することを示した。
