ポリアミンの生理作用解明と細胞内濃度調節機序

研究概要

1. プトレスシン取り込み系の基質結合蛋白質(PotF)のプトレスシン結合部位を、X線結晶構造解析及び変異PotFの活性測定より決定した。その結果、2分子のAsp、1分子のGlu、2分子のTrp、2分子のTyr及び1分子のSerがプトレスシンの結合に関与していることが明らかとなった。中でも、Glu185、Trp244、Asp247及びAsp278が最も重要であった。
2. 酵母のポリアミン輸送欠損株を用いて、ポリアミン輸送蛋白質遺伝子のクローニングに成功した。この遺伝子は12膜貫通領域を着する液胞膜上の蛋白質をコードし、この蛋白質は基質としてポリアミン(プトレスシン、スペルミジン、スペルミン)のみを認識した。
3. ポリアミン、特にスペルミンはNMDA受容体活性を脱分極時の促進、過分極時の阻害と二面的に調節している。この調節に関与しているアミノ酸の同定を試み、スペルミンによる活性促進にはGlu181、Glu185、Glo342、Trp608、Trp611、Ash616、Trp647及びAsp669が関与し、スベルミンによる活性阻害にはTrp563、Asn616、Glu621及びAsp669が関与していることを見出した。
4. ポリアミン、特にスペルミンが.ATP-Mg^<2+>と三者複合体を形成することをNMRを用いて証明した。この三者複合休形成により、ポリアミン輸送系のPotAのATPase活性が強く促進を受けることを見出した。また、protein kinaseAによる蚕白質のリン酸化も三者複合体形成により約2倍の促進を受けた。
