研究概要	アラキドン酸は通常生体膜の主要構成成分であるグリセロリン脂質にエステル結合した形で貯蔵されており、ホスホリパーゼA_2(PLA_2)の作用により適宜遊離され、更にシクロオキシゲナーゼ(COX)によって各種生理活性PGの共通の中間前駆体であるPGH_2に変換される。近年の脂質生化学の分野への分子生物学の浸透により、PLA_2には少なくとも14種類の分子種が同定され、従来PLA_2とCOXによって漠然と触媒されていると考えられてきた代謝系が、限られた分子種間同士の適切な機能連関によって厳密に制御されていることが明らかになってきた。我々は、アラキドン酸代謝系の律速酵素であるPLA_2の機能的役割分担とその調節機構について種々の細胞系を用いて検討を行い、即時的及び遅発的PG産生応答におけるPLA_2とその下流のCOX分子種間のカップリングの体系化を試みた。その結果、細胞質PLA_2(cPLA_2)と特定の分泌性PLA_2(sPLA_2-IIA、V)は即時、遅発両PG産生に関与するPLA_2分子種であること、このうちsPLA_2は周辺の細胞にPG産生応答を増幅する細胞間シグナル伝達因子として特有の機能を持つこと、Ca^<2+>の非依存性PLA_2(IPLA_2)とsPLA_2-XはPG産生には重要でなく、むしろリン脂質再構成に主に関与するPLA_2分子種であること、また二種のCOX分子種については、COX-1は即時応答でのみ、COX-2は即時、遅発両応答で機能すること、等の機能的役割分担を明らかにした。更に、特定のPLA_2分子種が選択的にPG産生を制御する分子機構に関しては、cPLA_2またはsPLA_2と相互作用するアダプター分子をそれぞれ同定し、これらを介した各酵素の細胞内局在性の変化がCOXとの機能連関の効率を規定していることが明らかとなった。
