薬物胆汁排泄機能の個人差の分子レベルでの解析

研究概要	肝臓は腎臓と並び、異物解毒において中心的な役割を果たす。我々は近年、特に肝細胞胆管側膜上に発現される一次性能動輸送担体(canalicular multispecific organic anion transporter;cMOAT/MRP2)の解析を進め、この輸送担体が種々抱合体を含む広範な有機アニオン系化合物を基質とすることを明らかとしてきた。更にcMOATにはホモログが存在する可能性も示唆されており、本年度はこのホモログのcDNAクローニング、機能解析を進めた。まず、MRP1および2のホモログとして、MRP3および6のcDNAフラグメントをcMOAT機能欠損ラット(Eisai hyperbilirubinemic rat(EHBR))より増幅し、その全長のcDNAクローニングを行った。更にMBP3に関してはLLC-PK1細胞をホストとした安定発現株を作成し、その膜ベシクルを用いることにより輸送機能を測定した。その結果、MRP3はグルクロン酸抱合体を基質とするものの、グルタチオン抱合体はpoor substrateである点においてMRP1および2とは大きく異なることが示された。一方、ヒトおよびラット肝臓より調製した胆管側膜ベシクル表用いた検討を進めたところ、グルタチオン抱合体に対してはヒトはラットの1/5程度の活性しか示さないのに対し、グルクロン酸抱合体に対しては両動物種でほぼ同等の活性が示された。ここで、MRP3は正常ラット肝では発現されないのに対してヒト肝での発現が観察されること、MRP3はグルクロン酸抱合体を特異的基質とすることを考慮すると、両動物種より調製した胆管側膜ベシクルにおける輸送能の差異はMRP3発現の有無により説明されうるものと考えられた。またラットおよびヒトMRP3は、誘導型であることが示され、排泄個人差を担う要因となりうるものと考えられた。
