研究概要	ヒトが生命活動を営む上で不可欠な血液の流動性は、血管内の抗血栓性機能を有する血漿因子と血管内表面の細胞膜受容体の分子間相互反応により維持されている。これまでに報告された幾つかの血液流動性維持機構の中でもプロテインC(PC)凝固制御系は特に重要であり、この制御系における血管内皮細胞上のPC受容体(endothelial protein C receptor:EPCR)、プロテインS(PS)受容体(EPSR)、トロンボモジュリン(TM)の構造と機能の解明はPC凝固制御系の分子機構の理解に不可欠である。申請者らは、これまでの研究で世界に先駆けてヒト血管内皮細胞からTMの遺伝子クローニングに成功し、現在、組み換えTM分子の抗血栓薬としての応用開発を行っており、また、ヒト血漿からPCインヒビター(PCI)を発見し、現在、その生理的・病理的役割の解明に向けた精力的な研究を行っている。こうした背景の下、本研究では、(1)ヒトEPCR遺伝子の全構造の解析、(2)未だ報告の無いEPSRの探索、単離および遺伝子クローニング、(3)EPCRやEPSRを介する細胞内シグナル伝達機構の解析、(4)先天性EPCR,TM異常症の探索と遺伝子構造解析などについて、分子細胞生物学的研究を行うことを目的とした。その結果、以下の成果を上げることができた。(1)ヒトEPCR遺伝子の全構造とその染色体局在部位を明らかにした。(2)血管内皮細胞上にEPSRの存在を示唆する結果を得、現在、その単離精製を行っている。(3)活性化PC(APC)による単球系細胞、血管内皮細胞、気道系上皮細胞での炎症性サイトカイン分泌の抑制作用を発見した。(4)APC作用により細胞内EPCRの分布が変動することを認めた。(5)PCIによる腫瘍細胞の増殖抑制、移転・浸潤の阻止作用のを明らかにした。(6)先天性EPCR欠損症およびTM欠損症の解析を行ったが、これまでのところ遺伝子異常者は見いだせなかった。
