蛋白質駆動ATPase SecAのダイナミックな動きを支えるSecY機能

研究概要

蛋白質が翻訳の場である細胞質から膜を越えて配置されるために必要な膜透過過程を補助する、大腸菌の細胞質膜における膜内在性トランスロケーター蛋白質複合体において、SecY,SecE,SecGの3者がどのように複合体を形成しているのかを、ヒスチジンタグをつけたSecYあるいはSecEとSecY-SecE相互作用を損なうsecY24変異体等を用いて解析した。その結果、SecYがこの複合体の中心に位置しており、SecE,SecGがそれぞれSecYと独立に結合していることを見いだした。膜透過の駆動力を提供するATPaseであるSecAは、それ自体が前駆体蛋白質を伴ってATP存在下で膜内に深く挿入しATP加水分解によって戻るといった、驚くべきダイナミックな動きによって蛋白質を膜内に送り込んでいる。このSecAのダイナミズムを損なわせるSecY変異体(SecY205)を同定し、それが膜透過チャネルの形状を変化させる結果、前駆体を伴うSecAのATPに依存した挿入を許さないものであることを提唱した。この欠損に打ち勝つサプレッサーSecA変異体を単離し、サプレッションの現象をin vitroで再現した。これらの結果から、SecYとSecAの特異的な相互作用の存在を初めて示し、またSecAの膜への挿入が実際に膜透過の駆動に重要であることを明らかにした。
