蛋白質駆動ATPase SecAのダイナミックな動きを支えるSecY機能

研究概要	蛋白質が翻訳の場である細胞質から膜を越えて配置されるために必要な膜透過過程を補助する、大腸菌の細胞質膜における膜内在性トランスロケーター蛋白質複合体の中心となるSecYと相互作用する蛋白質Sydの性質を利用して、SecYのSecEとの相互作用に関する新たな変異体を分離した。それらの解析の結果、SecYの残基240のSecEとの相互作用における重要性、残基249のSydとの相互作用における重要性を明らかにした。さらに、SecEとの相互作用が弱くなったSecY24変異型サブユニットをもつ膜透過装置に対して、Sydが強く阻害する事、その阻害はSecAの高親和性結合を喪失させることにあることを見いだした。一方、SecAの膜挿入を損なわせるSecY変異(SecY205)の欠損に打ち勝つサブレッサーSecA変異体の一つ(SecA36)がSecG欠失変異をも抑制することを見いだした。これら変異株からの反転膜小胞と精姑ecAを用いた解析により、SecGの役割はSecAの膜透過促進機能を助け、ATPを効率よく利用して蛋白質の動きを駆動するのことにあることを明らかにした。
