研究概要

タンパク質の構造および機能上の構成単位(構造ユニット)を実験的に決定する方法の開発を目指して、タンパク質の一次配列上の任意の位置を切断することを試みた。すなわち、構造遺伝子上に、ランダムに停止コドンと引き続いて開始コドンを挿入すれば、酵素タンパク質は、一次構造上次々と切断される。この切断遺伝子プールを持つ大腸菌から、酵素活性を持つもののみを選択すれば、どの位置で分断酵素が得られるかを決めることができる。この切断位置は、構造ユニットの切れ目であろうというのが本実験計画の狙いである。
本年度は、このための実験系の開発を行った。まず、トリプトファン合成酵素Aサブユニットをコードしている遺伝子trpAをモデルとして取り上げ、制限サイトを導入し、200-100残基毎のブロックに切り分けられるようにした。それぞれのブロックについて、間に停止コドンと開始コドンを挿入したリンカーによって繋ぎ配列を重複させる。一旦、この様なプラスミド(N末-(N-1)番目までのブロック-N番目のブロック-停止コドン-開始コドン-N番目のブロック-(N+1)番目以降のブロック-C末)について、停止・開始コドンを挟んで存在する二つのN番目のブロックの両方の内部をランダムに削り取り、分断配列のプールを作った。この変異体DNAを用いて転換された大腸菌(trpA)について、トリプトファンのない倍地で生育可能な株を選択した。
いくつかの分断酵素遺伝子が得られたが、そのうち解析されたものはN末-C81、P57-C末の構造を持つ分断酵素であった。酵素の性格などは次年度に解析する。
