TATAボックス結合因子TFIIDを中心とした真核細胞転写開始および調節機構の解析

研究概要	TATAボックス結合因子TFIIDは、真核細胞転写開始反応及び転写調節因子による転写活性化機構において中心的な役割を演ずる因子である。本年度においては、このTFIID因子の転写開始及び転写調節反応における機能と構造の関係を明らかにする目的で以下のことを明らかにした。 1.転写調節因子VP16によるTBP、CCG1との相互作用を介した転写活性化機構の解析転写調節因子VP16の転写活性化領域がTFIIDサブユニットCCG1と競合的にTBPとの相互作用の制御に関わることを明らかにし、転写活性化反応の一局面を明らかにした。 2.ヒトTAFII100をコードするcDNAの単離とサブユニット間相互作用の解析 TFIIDサブユニットTAFII100をコードするcDNAを単離し、WD40リピート構造を持つことを示した。発現した因子と他のTFIIDサブユニットとの分子間相互作用の解析を行った。 3.TATAボックス結合因子TFIID内ヒストンオクタマ-様構造モデルの提出 TFIIDサブユニット3種がヒストンH2B、H3、H4様構造を持ち得ることを示した。ヒストンフォールドモチーフ内に欠失が入るとこれらのサブユニット間の相互作用がなくなることを示した。 4.分裂酵母TATAボックス結合因子TFIIDサブユニットの遺伝学的解析分裂酵母からTFIIDサブユニットの単離を行った。WD40リピート構造を持つTFIIDサブユニットが生存に必須であること、また出芽酵母のものとは機能的に置き換えられないことを示した。 5.HIV転写に関与する新しいタイプのクロマチン因子の単離と解析 TFIIDと相互作用する因子として単離した因子の中に、Tat相互作用因子として得られた因子のひとつと一致するものがあった。この因子がヒストンアセチルトランスフェラーゼ活性を有する酵素であることを示した。
