肝細胞増殖因子(HGF)の医薬への応用に向けての活性制御因子の生体内動態の解析

研究概要

肝細胞増殖因子(HGF)は強力な肝再生因子であり、現在肝疾患の治療薬としての開発が進められている。HGFの活性は細胞外、細胞内の種々の因子により制御されている。本研究ではそれらの因子についてHGFの活性の制御との関係において解析し、以下の結果を得た。
1. 肝障害時にHGFを活性化するセリンプロテアーゼとしてHGF activator(HGFA)が同定されている。またこのHGFAの活性を阻害するインヒビターとして2種類のHGFA inhibitor(HAI)が精製され、両者は2つのKunitz型インヒビタードメインを持つことがわかっている。2型HAIについてそれぞれのKunitz型ドメインに変異を導入し、2型HAIの機能におけるKunitzドメインの役割を解析した。両方のドメインに変異を導入した2型HAIはまったく活性を持たなかったことから、HAI活性にKunitzドメインが関与していることが明らかになった。またN末側ドメインに変異を導入した場合に低い活性しか示さなかったことから、2型HAIの活性にはN末側Kunitzドメインが重要であることが明らかになった。
2. 細胞内においてHGFの活性を制御する新しいシグナル伝達分子として、Hrsとその結合蛋白質(Hbp)が同定されている。HrsとHbpは細胞内において複合体を形成し初期エンドソームに局在することがわかっている。HrsとHbpの複合体の機能を明らかにするために、HbpのHrs結合ドメインあるいはSH3ドメインを欠失するドミナントネガティブ体を発現し解析した。Hbpドミナントネガティブ体発現細胞では、増殖因子と受容体の細胞内分解が抑制された。したがってHrsとHbpの複合体は、HGFとその受容体が細胞内に内在化した後の分解を制御することによりHGFの活性制御に重要な役割を果たしていることが明らかになった。
