研究概要

本年度はRalGDSを介するRasの新しいシグナル伝達系について解析した。これまでに、翻訳後修飾を介して細胞膜に結合しているRasがRalGDSの細胞質から細胞膜へのトランスロケ-トを引き起こすことを明らかにしていたが、本研究によりCOS細胞においてRasがRalGDSを介してRalを活性化する際にRasとRalの両者の翻訳後修飾が必要であることが明らかになった。また、COS細胞においてEGFはRalGDS依存性にRalを活性化したが、この反応にもRalの翻訳後修飾は必要であった。さらに、リポソームを用いて、Ras/RalGDS/Ralから成るシグナル伝達系の再構成実験系を開発した。翻訳後修飾を受けたRasとRalがリポソームに取り込まれ、GTP型RasはRalGDS依存性にRalを活性化したが、GDP型RasはRalを活性化しなかった。RalGDSはRasの標的蛋白質結合部位変異体RasE37Gと特異的に結合し、Rafが特異的に結合するRasT35Sとは結合しなかった。RalGDSはRafと相乗的に作用してfos遺伝子の発現を促進した。また、RalGDSのホモログであるRGLも同様の性状を示した。Ralの標的蛋白質RalBP1はRalGDSと同様に翻訳後修飾を介して膜に結合しているRalにより細胞質から細胞膜へトランスロケ-トした。RalBP1と結合する新規蛋白質POB1(Partner of RalBP1)を単離したところ、POB1はRalBP1結合部位と異なる部位に結合して、Ral、RalBP1、POB1の三者は複合体を形成した。さらに、POB1はGrb2と結合し、EGF刺激によりチロシンリン酸化され、EGF受容体と複合体を形成した。以上の結果から、RalGDSはRasの下流でRafとは独立したシグナル伝達経路を構築して、Rafと相乗的に細胞機能を制御すると考えられた。さらに、その下流にはPOB1が存在して、RalからのシグナルとEGF受容体からのシグナルを直接受ける可能性があることが示唆された。
