研究概要

NO合成酵素およびキサンチン酸化酵素はフリーラジカルを生成する複合金属フラビン酵素である。本年度の経過と成果は以下のとおりである。1)NO合成酵素につき、大腸菌による発現系を開発し、現在全酵素(野性型および数種の変異型)およびヘムドメインの発現酵素を各種分光学的解析に供せられる量がとれるようになった。同時に安定化因子の主要物質がPhosphatidyletanolamine(PE)であることをphospholipase,PE特異的結合蛋白処理でその作用が消失することおよびTLC分析、TOF-massから調べた。種々の酵素につき物理化学的性質の解析を開始した。また天然に存在が知られる変異酵素(スプライシング異常により生じた変異酵素)を大腸菌による発現系により発現させ、その酵素がヘムを持っているが配位が異常であるために活性がないことなどを解明した。3)キサンチン酸化酵素のX-線結晶解析のデータ収集を終え、現在進行中である。またcDNA発現系を用いて、ニワトリ/ラットのキメラ酵素を作成、非ヘム鉄を欠落した変異酵素の作成を行い、その解析をおこなった。キメラ酵素を用いてこの酵素が脱水素酵素型から酸化酵素型への変換に関与する構造ドメインを同定した。2つの非ヘム鉄についてEPR、酸化還元電位および一次構造モチーフ上2つのタイプが知られているが、その同定を部位特異的変異法により作成した酵素を用いてその解析を行い同定に成功した。またモリブデン配位子であるイオウの転移に関与が推測されるイオウ転移酵素のcDNAクローニングとその反応機構および類似酵素ロダネ-ゼとの基質特異性の違いの理由を大腸菌の発現系を用いての解析をおこなった。また酵素の局在につき調べた。
