H^+-ATP合成酵素の溶液および固体多次元NMRによる構造・機能解析

研究概要	好熱性細菌由来のH^+-ATP合成酵素のTF_1ドメインのβサブユニット中に12個存在し、一次配列上比較的均等に分布しているTyr残基の芳香環の^1H-NMRシグナルのうちまだ帰属の明確でなかったシグナルについて2重の点突然変異を導入することにより帰属を行った。その結果、一つのアミノ酸残基が2本のシグナルを与えるものが最低3つあることが明らかになった。さらに、ヌクレオチドはこの二つの構造の一方にしか結合しないことも明らかになった。この際、試料調製に本研究で購入した振とう機およびインキュベートボックスを用いた。また、固体NMRでβサブユニット中のATPの構造解析を行うために、試料調製法をさらに検討したところ、トレハロース存在下で急速凍結法・再水和を行うとATP-TF_1βサブユニット複合体の^<31>P-NMRシグナルがさらにシャープになった。したがって、これは複合体構造を安定化させるものと考えられる。一方、Foの構成成分であるサブユニットcについては、再構成を考えて大腸菌の系を使うこととし、安定同位体ラベルされたアミノ酸を任意に組み込むことができるような化学的合成が可能であるとの目途をつけた。固体3次元異方性相関NMRを用いてC-13、N-15でフルラベルしたアミノ酸残基の側鎖の二面角を決定する方法論を開発した。これはα位とβ位のC-Hの双極子相互作用の相関を利用するもので、前者の双極子相互作用で展開した後にその^<13>Cの磁化をβ位の^<13>Cに移し、再びプロトンと双極子相互作用で展開する。この両者の相関スペクトルをさまざまな二面角のシミュレーションスペクトルと比較する。この際、^<13>Cα-^<13>Cβ間の磁化移動が非等法的になることを利用して高い精度で二面角お決定できる。二面角にととまらず、結合角、結合長も同時に決定できることが明らかになった。
