赤血球系転写因子の機能と発現制御機構の包括的な解析

研究概要	GATA-1遺伝子の生体での赤血球分化における役割を同定するため、GATA-1の赤血球特異的発現を担うIEプロモーターを破壊し、赤血球特異的にGATA-1遺伝子の破壊(ノックダウン)を行った。血球系でGATA-1遺伝子の破壊(ノックダウン)を行った。血球系でGATA-1遺伝子をほとんど発現していない固体は、胎生11.5-12.5日で致死であり、これは貧血によるものであると考えられた。詳しく解析してみると、赤血球の分化は前赤芽球で停止しており、はじめてGATA-1の生体内での作用点が明らかになった。また、β-ガラクトシダーゼをレポーターとしたトランスジェニックマウスの解析により、GATA-1の組織特異的発現に必要な上流域エンハンサーを同定した。このエンハンサーは、胎児期におけるGATA-1の発現には十分であるが成人型造血におけるGATA-1の発現には十分でなかった。成人型赤血球細胞でのGATA-1の発現にはさらに第一イントロンが必要であることが明らかになった。 GATA2の発現を調べている過程で、2つの第一エキソンを発見した。GATA2遺伝子は血球以外の組織にもひろく発現しているが、これはIGエキソンに由来している。今回新たに発見したISエキソンは血球特異的に発現されていることがわかった。 Nrf2遺伝子破壊マウスでは、血球分化には異常はなく、新電子性物質による第二相異物代謝酵素群の誘導に欠失がみられた。Nrf2のN末端には活性を負に制御するドメインが存在しており、このドメインを介して、誘導のシグナルに反応していることが明らかになった。このドメインには、細胞骨格に相互作用する因子Keaqp1が相互作用し、Nrf2の活性を負に制御している。 NF-E2の小サブユニットであるMafKの発現も、同様に2つの組織特異的なプロモーターによりなされている。造血細胞系におけるMarK遺伝子座のDNase高感受性領域を調べ、新たに胎児肝造血部位においてMafKの発現に必須であるエンハンサーを発見した。
