研究概要

1.アブラナ科植物の自家不和合性認識反応における花粉因子の探索(磯貝・高山)
アブラナ科植物の自家不和合性では、これまで、S遺伝子産物としては柱頭側の因子として、Sレセプターキナーゼ(SRK)とS糖タンパク質(SLG)が明らかとなっていたが、花粉側の因子は、不明であった。本研究では、SLGと相互作用をする花粉抽出物の精製、蛍光ディファレンシャルディスプレーを用いたS遺伝子型で異なる花粉mRNAの探索を通じて、花粉因子の候補分子を見いだし、その分子のS遺伝子間での多様性、SRKなどとのゲノム上での位置関係、花粉や、葯での発現様式を解析し、さらに、この遺伝性の形質転換実験、および、柱頭上での花粉の吸水反応を指標とした生物検定系によって、これが花粉因子であることを確定することが出来た。便宜的にSP11と命名した花粉因子は、システイン8個を含む分子量約70000のタンパク質で、アミノ酸組成はS遺伝子毎に大きく変異していたが、シグナルペプチド部分は高く保存されていた。
2.植物病原菌のフラジェリンと相互作用する因子の探索(蔡)
単子葉植物を宿主とする植物病原菌Pseudomonas avenaeのうち、イネを宿主としない菌株は、細菌を認識して、抵抗性反応を示す。まず、この抵抗反応はある種の過敏感反応であることを細胞生物学的に明らかにした。さらに、この抵抗反応を誘導する細菌側の因子が、鞭毛を構成するフラジェリンであることを、フラジェリンタンパク質の精製及び、フラジェリン欠損菌株の作成によって確証した。また、菌株ごとのフラジェリン分子の差異が、分子の中央部分に集中していることをあきらかにした。
