血小板放出(PPF)因子の同定と血小板産生の分子機序

研究概要	TPOによって巨核球が成熟分化することが明らかになったが、多倍体化した巨核球から血小板が放出される分子機序に関しては不明のことが多い。そこで骨髄細胞から巨核球を単離しTPOを投与し成熟巨核球を得、血小板放出(PPF)のin vitroでのアッセイ方法を確立した。PPFにはTPOを始め既知の因子は全く関与していないことが判明した。そこでPPFを促進する外来因子の存在の可能性を調べた。その結果、PPFが促進される因子が血小板減少症患者血しょうに存在することを見い出した。また、ある特定細胞上清にも同じ分子量分画にPPF促進活性が見い出され精製を試みた。この粗精製品はマウスに投与すると24-36時間後に血小板を顕著に増加させることが出来た。TPOは5-7日後に血小板を増加させるのに対比して即応性が高かった。現在この因子の遺伝子を単離中である。また多倍体化する巨核球を細胞生物学的に解析した結果、株化された細胞で言われているような細胞分裂(M期)をスキップしてDNA合成のみが反復されるメカニズムではなく、巨核球はM期に入りprophase、metaphase、anaphase Aまでは進行するものの、姉妹染色体が極まで充分引かれないために核膜が複製した染色体を全て包括してしまう為に起きることが判明した。
