プロラクチンによる両生類精原細胞のアポトーシス誘起機構

研究概要	我々は、ヒツジプロラクチンをイモリ腹腔に注射すると、第一精母細胞に分化する直前の精原細胞が細胞死を起こし、FSHによってレスキューされることを見いだした。本研究は、プロラクチン受容体がどの細胞に存在するのか、FSHの作用とプロラクチンの作用はどうして拮抗するのか、プロラクチンによってどうして減数分裂開始直前の精原細胞だけがアポトーシスを起こすのか、プロラクチンによる精原細胞のアポトーシスの生理的意義は何か、を明らかにすることを目的とする。本年度は、次のことを明らかにした。 (1) 低温下での精原細胞のアポトーシス: 低温下にイモリを置くと精原細胞が死ぬ理由を調べるために、イモリを22℃〜8℃で飼育し、血中のプロラクチン濃度を測定したところ、8℃や12℃では22℃や18℃に比べて約3倍高いことが明らかになった。また、8℃においてイモリプロラクチン抗体を注射したところ3日間は精原細胞のアポトーシスを回避した。この結果、低温下で精原細胞が死ぬのは、イモリ下垂体から血中へのプロラクチンの分泌量が増すためであることを証明した。 (2) イモリプロラクチン受容体遺伝子の発現: クローニングしたプロラクチン受容体遺伝子全長をCOS細胞に発現させるとその膜分画にプロラクチンが結合することから、機能的受容体であることを証明した。イモリプロラクチン受容体遺伝子の精巣における発現をRT-PCRで調べたところ、ステージ特異性は見られなかった。また、同遺伝子の発現はセルトリ細胞の方が高いものの生殖細胞でも発現が見られた。
